A Synch Nexus was a state during which the memories of an individual and his ancestors converged.

In the year 2012, Desmond Miles fell into a coma after stabbing his fellow Assassin, Lucy Stillman, under the influence of Juno. Upon being plugged into the Animus and being sent to the Black Room, Desmond met the visual representation ofSubject 16. Subject 16 eventually told Desmond that the only way to escape from the Animus, was to achieve a snych nexus. After being told this, Desmond began to relive the later memories of his ancestor, Ezio Auditore da Firenze.

Upon completing the final memories that he had not relived of both Ezio and Altaïr, Desmond was pulled into the synch nexus. Once there, Jupiter, one of Those Who Came Before spoke to Desmond, revealing all that humanity and the First Civilization has faced up to that point, and may face in the future.

After returning back to the Black Room, the Animus scheduled Desmond and Subject 16 for deletion, though 16 pushed Desmond out of the way, saving his life, but sacrificing his own. Desmond then awoke from his coma, telling the other Assassins that he knew where to go.
