Over the course of his training to become an Assassin, Desmond Miles had various conversations with different people, along with other Assassins and major characters interacting with each other throughout modern times.
Assassin's Creed
While being held captive at Abstergo Industries, Desmond secretly had multiple conversations with Lucy Stillman, Dr. Vidic's assistant. Before each Animus session, he would also have conversations with Vidic, who would discuss certain details of Abstergo's goals.

Desmond laughed.
Lucy scolded him back for his prank.
Afterwards, Lucy jumped down, and Desmond began to see more figures through the Bleeding effect.
Desmond heard a conversation between Claudia Auditore and Ezio, due to the Bleeding effect.
The ghostly forms disappeared.
Desmond walked toward a lift, and pulled Lucy close to him.
Desmond kicked back the lever on the lift, and the two of them rocketed upwards to a ledge.
They began to freerun through the tunnel.
Lucy replied back jokingly.
Desmond reassured Lucy.
They both came to another gate and lever.
Desmond feigned innocence.

Desmond entered Santa Maria Aracoeli through a secret entrance, while Lucy, Shaun and Rebecca waited at the door outside.
Desmond had a vision of Ezio climbing up the altar.
Desmond activated what would turn out to be a wall lever, and multiple poles shifted out of the ceiling, constructing a series of free-running obstacles.
Desmond activated another switch, and a ladder came out of the wall.
Desmond eventually activated a switch on the highest platform, and a pedestal came out of floor at the front of the church.
Desmond moved his hand above the pedestal, which activated into a lift on sensing his First Civilization genes, that took the Assassins underground.
The lift stopped, and the entrance to Juno's Temple could be seen.
The Assassins approached the door.
The door opened.
Desmond activated the pedestal, and multiple platforms and beams came out from the floor and walls.
Desmond activated all of the switches, and stairs to the central platform came out of the floor.
The Assassins approached the Apple of Eden.

Desmond woke up in the Black Room, before taking some steps forward to adjust to his surroundings. Meanwhile, the digital construct of Clay Kaczmarek (Subject 16) initialized nearby.

Desmond explored the Temple looking for a socket for the battery he'd just acquired. Along the way, he encountered Juno, who explained the solutions the First Civilization had tried to save the world from the First Disaster.

Desmond soon found a socket and plugged the power source into it. When this was done, he returned down below and talked to Rebecca.
Rebecca's voice broke as she related what happened next.
Desmond then spoke to his father next. William first tried to apologize for the earlier punch, then make peace for Desmond's rough childhood.
Desmond then asked his father about whether William had ever looked for him.
Desmond then lamented the fact that more often than not, the Assassins keep defecting to the Templars than vice-versa.
Desmond then made his way over to Shaun, who wanted to use the Animus to go back to the beginning.
Shaun then launching into seeing the American Revolution from a decidedly British perspective.
Shaun then wondered how many other First Civilization vaults there were still to find.

The team proceeded to make their way towards the ending of the path leading to a round device.
Desmond was shown a vision of what would happen to the world if he refused to use the device.
Desmond, Juno and Minerva then continued their conversation.
Desmond touched the pedestal, killing himself instantaneously but saving most of the world in the process.
