The Ise are an ancient family of warriors who served various administrative functions for the Ashikaga shogunate. Several members of the clan left treaties on administration and the proper conduct of warriors at the shōgun's court. Although very young, Sadatame was put in the service of the 14th shogun Ashikaga Yoshihide. However, upon the sudden death of the shōgun in 1568 Sadatame was, for a time, considered part of the unreliable officials by the new shōgun Yoshiaki who had formerly served his cousin. As a result, he was sidelined for a time. Yet, his qualities, as well as those of his younger brother Sadaoki, were eventually recognized. Sadatame was integrated into the otomoshū, the shōgun's commensals, those who served him daily.

In 1573, relations between shōgun Yoshiaki and Oda Nobunaga deteriorated. Sadatame then fought at the Nijo Palace in Kyoto for his lord, but Yoshiaki was defeated and expelled from Kyoto. Nobunaga then ended the Ashikaga shogunate. Nevertheless, he pardoned Sadatame and his brother Sadaoki, probably due to their administrative skills. Sadatame then worked in the offices of Nobunaga's new regime as his vassal. Having taken ill, Sadatame resigned from his duties in 1581, but resumed his activities with Hideyoshi for a while before retiring again due to his health. He spent the last part of his life writing treaties on horsemanship and warrior traditions and his sons entered the service of the Tokugawa administration.
