A merchant defeats Donato in a race. Mancini responds angrily to his defeat by destroying the merchant's stand and threatening his life. This is made known to the Assassin Ezio Auditore da Firenze who decides to kill Donato. He locates him on horseback at the [[Circus Maximus]], surrounded by his guards. Ezio proceeds to only kill Donato, sparing the lives of his guards.

Donato rode well and fairly in the competition, despite the interference of one of Cesare's officers, [[Teodor Viscardi]], who charged him in the middle of the race and attempted to attack him with a blade. Donato only dodged the thrust and spurred his horse forward to win, claiming his victory before a silent audience.<ref name="ACPL"/>
[[Fiora Cavazza]], a Templar agent of [[Cesare Borgia]], came to Donato and asked him to lose the upcoming horse race against Cesare. He refused to and barely won the race but with dire consequences. He was severely punished by Cesare but was still employed as an agent.

Donato Mancini (1473 – 1503) was a military captain of the Papal Army under the Borgia, and a member of the Templar Order. While under the direct employ of Cesare Borgia, Donato was often described as a confident and capable horseman.

In 2012, his genetic memories were used as an Animi Avatar by the Templar company Abstergo Industries, in the first stage of training for their Animus recruits, under the title of the Captain.

In 1502, during a private horse race held by the Borgia, Fiora Cavazza was sent to Donato to either bribe or threaten him into allowing Cesare to win.[1]
