I think your general theory is right. Adam and Eve ( in the game) were the first humans to evolve so that they developed resistance to the mind control from the pieces. In doing so they saw the injustice and stole a piece to escape with it. They started the human revolution and due to the numbers they were able to break free. They had two sons, Cain and Able and it was said that Cain was the first Templar and his "sign" became the Templars. However, I do not think that Able was the first assassin. It also has not been shown that the Templars are all resilient to the pieces of eden's mind control. I believe the "ones that came before" mated with some humans and the offspring from those were born impervious to the effects and this is where the assassin bloodline began. It makes more sense if the ancestors were complete "half-breeds" and were able to pass the traits along than to say that ALL the assassins came from adam and eve because by now many other humans would have naturally evolved without those neurons (having not been needing them for thousands of years). This would also mean that the Templars are not exactly immune to the effects, even though they still probably have diminished effects. Then again, there was that letter from a witness at the Colorado International Airport who obviously wasn't affected by it either so maybe some of the general public have evolved too. Its very interesting how they've been able to tie so much of it in with real history. I love it.
