Subject 16 is not directly related to Desmond. He is however also related to both Ezio and Altair. The most popular theory is that Altair is related to Adam and Eve. Their children chose opposite sides. One became an Assassin and the other became a Templar (the very first). Adam and Eve obviously had the resistance to the Pieces of Eden and passed this down through their children (although their only surviving child was the Templar one so how Altair's family became Assassins is not known) all the way to Ezio and he passed it on to every one of his descendants down to Desmond and Subject 16 (although whether he has the resistance is not known)
