As far as the story with Cain and Abel goes, i think it would be correct to assume Cain was the first Templar and Abel the first Assassin. If i am correct, in the BIBLE it states that Cain killed Abel out of jealousy. But he could have killed Abel because he was commanded to do so by "Those Who Have Gone Before". Thus making him, just as the Templars are now, loyal to the Father of Understanding, or Those who have gone before. Plus, it is possible that the "Mark of Cain" was something making him resiliant to the Pieces if Eden's effects, because he was obedient to them and wouldn't need to be controlled. From then on he could pass it down to other Templars, and so on.
