In regards to their prime, I mean their prime in terms of skill & fitness combined. So basically whenever they were at their best. I would say Ezio was at his peak in Brotherhood.

Also, to make it more fair, let's say that all of them are well-informed of the others' technology. That doesn't mean that they are aware of their skills though, just that Ezio & Altair etc are aware of guns & so on.
