I don't believe Ubisoft would put major characters in their multiplayer portion of Assassin's Creed.

Vali cel Tradat (Sentinel) and Shakulu (Renegade) worked in MP because while they were part of the main story, they weren't major antagonists. Vali was basically the equivalent of a Master Assassin/Den Master mission that was incorporated into the game's main events, and Shahkulu's purpose was to be the bodyguard for one of the bigger antagonists, Manuel Palaiologos.
