The animus reads the genetic memories of the one using the device, as an example; it is not possible to read your brother 's DNA because it isn't "into you" (just like you can't inherit your kid's disease, that 's why it 's called "reliving", you can't relive something who "didn't live yet"), so it is not possible that Desmond will be able to just read 16's memories, and it also won't be possible for a random animus user not related to the one who did put his/her memories in to read the memories, I think it works like blood transmisswion or something...
