I have no opinion on this, but it could be that Subject 16 set up the Glyph locations as an "introductory" task of sorts, we already know that at the beginning of AC:Brotherhood it was inside of the Glyph files the the Rift coordinates were found, so maybe Subject 16 planned it so that after the Glyphs were completed, the team would be lead to Ezio's memories in Rome, where by completing they would have found all of the Rifts and the files they contained, which would have perhaps "activated", or "awakened" the electronic "consciousness" hidden inside of the animus. Just a thought.
