QOTW - I liked all the "atmospherical" jokes that took place. There were more in later games, though, but just examples:

AC2: Cynical Maria and brash Ezio: "I have plenty of outlets!" "I meant. Beside. Vaginas!" (yes, with pauses)

ACB: Lucky Niccolo: "They say, Jesus Christ resurrected three days earlier."

ACR: Funny Yusuf explaining bombs: "One wrong move and... Hehe, you should've seen your face!"

AC3: Rational Achilles: "What is your name?" "Ratonhnhake:ton" "I wouldn't try to pronounce that!"

And also little details, like scold on Connor's face, when Paul Revere slaps him on the shoulder, "since Indians have such a high respect for one's body".

These details make the game awesome.
