Well assuming the glyphs are a literal tie in with the Adam and Eve memory, you would have a single Templar. Technically you would have one Templar out of that joining, though we don't know if the first civilization lasted long enough for that.

For some reason Assassins tended to be hybrids, that could be explained in a first civilization game, but we don't know any order older than the Persian Order that the Assassin Darius belonged to.
