Their personalities is mostly what I mean when I say they are unique. Of course the share similar lives given their ties to the Assassins and the structure of the game series. But each have separate motivations, demeanors, beliefs, and style. Though I will admit I think that Connor is very similar to Altair and from what I have seen of Edward he seems more like Ezio.
