It's possible. IMA GIFTEHD FREEFRUNNERH GURLLLL SHOOOOO. In the first part, run all the way up into the basilica, find the cardinal and just follow him around while blending in with the others (Pretty obvious, and now I'll just get to the Points of Interest) First obstacle: (The annoying place where the camera betrays you and goes to it's own spot) Climb up the wall and scale towards the left until the camera is back to free mode again, fast as you can, then back jump. Second: After you climb all the way to the top of the tower thing, two windows/doorways right? Go to the nearest (The one facing you when you're all at the top. Before you is a spire with a Borgia flag atop, Back jump from the wall opposite and ledge grab it. Then jump from the spire onto the roof (This saves about 3-4 seconds from the usual route but may cost points. Third: (I suggest you practice with the map first and memorize the obstacle course throughout) then you can go through at full speed. Fourth: Once you catch up with the cardinal, grab him, and throw him towards the railing (Cutscene occurs, Ezio grabs the key, drops him blah blah blah. Saves more time) Then run towards the view point, DONT JUMP, but drop onto a pole beneath, and you can get the Borgia flag AND still Leap of Faith into the well thing. When you open the chest in the lair, the timer will stop anyway. I managed to do the mission in under 7 minutes with these tactics.
