About the questions well hre it is : The vault will be closed forever because even the pope can't open it. The romans will suffer, Caterina will nbe imprisoned for a much longer time, And The ottoman assassins can't deal with massive amount of Templar attacks, Yusuf & Suleiman Won't be helped, and finally no Flavia and Marcello And no Desmond...
