The Basilica di San Marco is a large cathedral church located in the San Marco district of Venice. It was constructed in Byzantine style in the 11th century. It is located next to the Palazzo Ducale, another prominent structure.
In the Game it is a heavily guarded building, you enter it once with leonardo's flying machine. However, That Isn't the only way to enter it. you learn the way in the quest where you scout the building.
There is also an Assassin's Tomb here that protects Amunet's Seal. Once you enter the Basilica via the skull-switch, Ezio attempts to pull four levers that are situated at the ceilings of the four halls of the inside. Each lever rotates a circular shape of the mural in the center of the basilica that, once all levers have been pulled, forms the Assassins Emblem that causes the mural to retract onto the floor revealing Amunet's Tomb. St Mark's Bascilica is a restriced area in game and is surounded by many guards.
