The surprise will be the modern day story and the events surrounding Desmond, the Assassins and key relationships that will be introduced at the beginning. I also expect there to be either a portable animus or Desmond can now partially control the bleeding effect through meditation.
Logically the story has to begin with Desmond and how and why he needs to revisit Connors memories.
