My theory is that the surprise will be the modern day story and the events surrounding Desmond, the Assassins and key relationships that will be introduced at the beginning. I also expect there to be either a portable animus or Desmond can now partially control the bleeding effect through meditation (to offset the negative effects we saw happen to 16).

Logically the story has to begin with Desmond and how and why he needs to revisit Connors memories. (Unless they do something completely radical and unexpected and make the Connor angle be an ancestor relieved by someone else entirely). Like Guarddog pointed out, it may be an unexpected assassination but I feel (whatever it is) that Ubisoft will start the beginning with a bang, something that throws us into the story, the sense of urgency that warrants a story about the earth being destroyed in the less then a few weeks (in game) while having to deal with the Templars.
