Gameplay
Plot
After forcefully stabbing Lucy Stillman at the command of Juno, Assassin Desmond Miles entered a critical coma, and is being kept alive by the Animus (put there by William Miles). William and Rebecca Crane prepared to leave for New York with Desmond while Shaun Hastings stayed in Rome and attended Lucy's funeral. Desmond's brain remained active while in the Animus, though the team watching over him was not able to record anything while he was in the machine.
Trapped within the Animus' Black Room, Desmond's consciousness awoke at Animus Island, and was greeted by Subject 16, whose consciousness has been alive and imprisoned within the Animus' software. 16 explained that due to Desmond's prolonged time within the Animus, his mind was now meshed with the memories of his ancestor; all their memories are starting to interfere with his own, as well as his former ancestor, Altaïr. In order to free himself from his coma, Desmond must relive the last greatest moments of Ezio until nothing was left for him (reaching the Synch Nexus). This will separate his mind from the memories of both his ancestors; Ezio, and Altaïr Ibn-La'Ahad.
Desmond relived Ezio's life once again, during 1511. Ezio has set out to uncover the secret library left behind by Altaïr in Masyaf after discovering a parchment from his father. There in Masyaf, he was greeted by a large platoon of Byzantine Templars, who successfully captured him after a fight. Brought upon the tower of Masyaf's Assassin fortress, Ezio was to be hung by the Byzantine captain Leandros, but successfully escaped. Navigating through the ruined fortress, Ezio happened upon a secret entryway to Altaïr's library.
Ezio soon discovered that the door was locked, and can only be opened by special keys made by Altaïr. Learning that the captain Leandros retrieved the journal of Niccolò Polo, which would supposedly lead him to the keys' location. Making his way outside the fortress, Ezio followed Leandros and chased after him through Masyaf's countryside, leading him to a nearby remote village. There, he was able to kill Leandros (who spoke of a Grand Temple being sought after by the Templars), and retrieved the journal. Learning from the journal that the keys were hidden in the city of Constantinople, Ezio left Syria, and sailed for the great city.
Arriving at the city with the Ottoman Prince Suleiman I (unaware of his name or title), Ezio arrived at the city of Constantinople, and was greeted by the Turkish Assassin leader, Yusuf Tazim. After a brief tour of the city, some time training with the Ottoman's trademark weapon, and assisting in the protection of one of the Assassins' strongholds, Ezio set out to find the Polos' old trading post. He was directed by Yusuf to an old bookshop owned by the Venetian Sofia Sartor. Inspecting the shop, Ezio uncovered an entrance to the old Yerebatan Cistern, and managed to retrieve both a map and one of the keys. Disclosing what he found to Sofia, Ezio agreed to work with her to uncover the location of the old books indicated in the map that would lead him to the other keys, while keeping information about his affiliation and workload a secret.
While helping the Ottoman Assassins, Ezio learned from Yusuf that the Byzantines planned to assassinate Prince Suleiman during a banquet at Topkapi Palace. Infiltrating the palace disguised as minstrels, Ezio and Yusuf saved Suleiman successfully from the Byzantines. The Prince was grateful for Ezio's help, furious that his own personal guard could not protect him. Suleiman summoned the Janissary Captain Tarik Barleti to Topkapi for a meeting with him and his uncle Prince Ahmet, successor to Sultan Bayezid II. Talking with Ezio in private, Suleiman asked the Assassin to help him in his investigation with the Byzantine's attack, starting by eavesdropping on the Princes' meeting with Tarik. Ezio agreed, and listened to their meeting while hiding in a hidden room.
Ezio believed that Tarik was the prime suspect, and proceeds to tail him in order to prove his suspicions. Starting from the Grand Bazaar, Ezio tailed Tarik all the way to the arsenal of the city, where he saw him negotiating with Byzantines. With the arsenal gates locked out by the Janissaries, Ezio met with Yusuf, and thought of ways on how to open the gates. Utilizing the nearby heralds, Ezio hoped to incite the civilians into causing a riot that could break through the gates. Despite Yusuf's disapproval and the Janissaries' attempt in containing the crowd, the rioting mob broke through the gates. Ezio entered the arsenal and located the warehouse where Tarik's contact were. Hearing the name Manuel Palaiologos from one of the Byzantines, Ezio proceeded to inspect the crates given by Tarik; crates that contained several firearms. Hurriedly leaving the arsenal, Ezio prepared to report back to Suleiman.
Disclosing what he learned about Tarik with Suleiman, Ezio was to extract information from Tarik about where the firearms were headed, and then kill him. Assassinating a lone Janissary, Ezio stripped disguised himself as one of the Janissaries and proceeded to infiltrate their camp. From there, he went deeper until he saw Tarik, who was discussing his arrangement with the Byzantines with one of his own men. After learning that the firearms have made it to Cappadocia, Ezio assassinated Tarik. During his last words, Tarik confessed that he was scheming an ambush for the remaining Templars at their home territory; he sent some of his subordinates to infiltrate the Templar ranks and report to him about their activities.
