Sima and anon are correct here. They were destined to said to Cyprus before Ezio assassinated them, and Rodrigo never went himself. Remember the All Things Come to He Who Waits in AC II when the ship returned to Venice with the Apple? And it was given to a courier to deliver to Rodrigo? Ezio tailed the courier before killing him and taking his armor. Then in the next Play Along, he posed as the courier and "delivered" the Apple to Rodrigo. And that's when he fought him alongside the other Assassins.
