The Pythagorean Vault was a comparitively small chamber constructed by Those Who Came Before and later incorporated into the Temple of Pythagoras. The chamber was largely hollow, save for a small pedestal located in the center of the main chamber on a raised base.

In 1506, the Vault was visited by Ezio Auditore da Firenze and Leonardo da Vinci during their exploration of the Pythagorean temple.

Inside, they were confronted by a mysterious sequence of numbers and letters: 43, 39, 19, N, 75, 27, 42, W. Though these meant absolutely nothing to Leonardo, a modern audience would recognize them as coordinates, and Ezio realized that they were yet more "words are not meant for [him]" - a message similar to the one he received from Minerva for "Desmond". Ezio merely told Leonardo that the message was not meant for either of them, perhaps tiring of his friend's loose tongue.
