Tsuyu was saved from dire circumstances as a young girl by an Assassin, a foreigner named Catarribera, and trained in the Creed. She became the ultimate weapon—the consummate Assassin.

She is the Assassin who indoctrinated Iga's greatest shinobi into the Creed; Tsuyu trained Momochi Sandayu, Hattori Hanzō, and Fujibayashi Nagato. She built the Kakushiba Ikki—the League of the Hidden Blade—and directed her recruits to protect the regalia, entrusted to the Assassins by the emperor years prior.

While in Iga, Tsuyu fell in love with Nagato and together, they had their daughter, Naoe. But after Hanzō betrayed the League and the regalia were lost, Tsuyu reluctantly left Iga and her family, faked her death, and attempted to recover the lost regalia according to her sacred duty.

During Tsuyu's hunt for the Sword, she was captured by a TemplarKimura Yukari—and imprisoned on Awaji for years as Yukari attempted to extract information about where Tsuyu had hidden the Sword. But Tsuyu would not break.

When Naoe is reunited with Tsuyu on Awaji, having suffered years of imprisonment and harsh treatment, she finds Tsuyu is not physically capable of being the warrior and commander that she once was—but Tsuyu's mind has trouble accepting her new reality.
