Prospero Colonna (1452 – 1523) was an Italian condottiero in the service of the Papal States and the Holy Roman Empire during the Italian Wars.

In 1500, Prospero and his cousin Fabrizio Colonna were captured by Cesare Borgia. While they were being transferred from one prison to the next, they were rescued by Ezio Auditore da Firenze and the three of them fought through a battalion of guards. After having won the fight, the Colonna cousins promised to continue their fight against the Borgia.
