The Followers of Romulus were adherents of Romulus, the mythical founder of Rome. According to legend, he and his brother Remus were raised by a wolf, and as a result, were said to be half wolf and half man. In tribute to this, the Followers' attire included a skinned wolf pelt. In Rome of the 16^th century, the Master Assassin Ezio Auditore da Firenze was able to enter six of their lairs to find out more about them, and to obtain their scrolls, which were located at shrines inside each of the six locations.

In Assassin's Creed: Brotherhood, the Followers of Romulus are portrayed as powerfully-built, rather brutish men covered in wolfskin pelts, who attack viciously in large packs. They are armed with various short blades and throwing knives, and tend to growl and snarl when in a fight. When they were killed, they howled like a wolf. It was possible that they were adept freerunners, as the Lairs that they occupied requires such skills.

They are supported by the Borgia in their actions, and are mainly used to cause fear in the civilians to send them into the arms of the church. However, the Followers are used and misguided, with their leaders commanding them in the name of the god-like figure of Romulus, despite not believing in his existence themselves.

By exploring and completing all six of their lairs, Ezio is able to obtain six keys to unlock their secret armor, the Armor of Brutus, one that is stronger than the armor Ezio can purchase from a Blacksmith, except for the Seusenhofer armor. The armor is also accompanied bt the Dagger of Brutus, a dagger with five stars in each category. Each key is accompanied by a scroll, which, when combined, tell the story of how Brutus and other followers plotted the murder of Julius Caesar. In the scrolls, it is written that Brutus planned to hold the first meeting under the Colosseum. There, he found the Vault containing a Piece of Eden, and used it to plot the assassination. The fact that he was able to open the Vault, and successfully plan and conduct the killing of Julius Caesar imply that he was an adept Assassin.
