The guild was formed in 1500 by [[Niccolò Machiavelli]], who had moved to [[Rome]] to directly confront the [[Templars]] in their headquarters. Machiavelli got in contact with [[Fabio Orsini]], who lent an unused storeroom on [[Tiber Island]] to him, which became the guild's location. Soon after its formation, [[Ezio Auditore da Firenze]] moved to Rome as well, after his hometown of [[Monteriggioni]] was [[Fall of Monteriggioni|ambushed]] by the Borgia; the leading body of the Templars.

Successfully forming a Guild, Ezio Auditore established contact with the Roman Underground (the [[Courtesan]]s, [[Thieves]] and [[Mercenaries]]) and restored each faction, successfully providing the Guild with powerful allies and resources.
 
In 1501, however, Ezio concluded the Guild must expand in numbers and thus began recruiting several civilians being bullied by the Borgia and training them as [[Apprentices]] to eventually become Assassins themselves. Sending the recruits across Europe, they completed various assignments, directly helping the Guilds in other European countries and forming one in [[Constantinople]] after many failed attempts in the past.
 
By 1503 the Roman Assassin Guild had infiltrated the Borgia's ranks, gained several powerful allies, and resources. In addition to killing several key generals and right-hand men of [[Cesare Borgia]], Ezio, now [[Grand Master of the Assassin's Order|the Mentor]], intended to assassinate both Cesare and [[Rodrigo Borgia]]. In the aftermath, Rodrigo died by Cesare's hand and the Assassins had successfully retrieved [[The Apple of Eden|the Apple of Eden]]. In the following months, the Roman Guild constantly worked on weakening Cesare's remaining supporters until his eventual arrest.
 
Ending the Borgia tyranny in Rome, the Guild continued its activities in Rome and throughout Europe aswell.
