The number 72 played a significant role to the modern day Assassins. Most notably, it was used as the password to gain access to the Colosseum Vault during October 2012.

History
As evident in the Scrolls of Romulus, the earliest known user of the number 72 as the password to the Vault was Marcus Junius Brutus, circa 45 B.C.E. The next known person to open the Vault was Ezio Auditore da Firenze in 1507, to seal the Apple of Eden within.

Due to Desmond Miles' vision of Ezio in the Sanctuary, it is known that Ezio later returned to the Villa Auditore to leave behind the clues to the password that would allow the modern-day Assassins to access the Vault, and obtain the Apple.

Ezio left the clue on the Sanctuary staircase, just behind the bookcase in Mario's study. It consisted of an etching of an equilateral triangle with nine other equilaterals within it – the same marking found on the door of the Colosseum Vault – and the numbers 1419, 1420 and 1421, which were only visible to those who possessed the gift of Eagle Vision.

Modern-day discovery
When the group of Assassins first arrived at Monteriggioni, and gained access to the Sanctuary, Desmond noticed and read out the numbers written into the wall. Shaun Hastings posited them to indicate years, though he could not think of their significance to each other.

After discovering the location of the Apple through the use of the Animus, the Assassins pondered on how to access the Vault under the Colosseo. Rebecca Crane discovered that the door required a password, but a power failure cut short their attempts to discover it.

As the Assassins waited in the dark and contemplated their next move, Desmond remembered the markings, and pointed them out. Shaun finally discovered the significance of the numbers and explained them to the rest of the team; pointing out the 72 names of God found in Exodus chapter 14, verses 19 through 21.

Having obtained the password, the Assassins left the Sanctuary, and headed for the Colosseo. As Desmond made his way through the ruins of the Colosseum and the path of the Lair of Romulus underneath, Lucy Stillman realized that the current date, October 10, was exactly 72 days away from the Abstergo Industries satellite launch in December 21.

Though she wondered aloud if this was more than a coincidence, Desmond dismissed her unease, stating that; "Someone probably wanted to make sure we get the password right."

The team eventually arrived at Juno's Temple, where the door to the Vault was located. After briefly wondering if the door would understand English, Desmond spoke "72" aloud, thus opening the Vault and allowing the Assassins to enter.

As Desmond activated the Apple, Juno told him that his journey would begin on the 72nd day before the awakening.

Trivia
*Both Rodrigo Borgia and Maria Auditore died at the age of 72.
*Shaun Hastings mentions that the construction on the Colosseum started in the year 72 C.E.
*William Miles mentions in an e-mail to Shaun that the team in Osaka was scheduled to make contact with headquarters in 72 hours, but that they went "dark."
*Giovanni Mocenigo was the 72nd Doge of Venice.
*At the end of Assassin's Creed: Brotherhood, Shaun mentions that a Phrygian Cap and a Masonic Eye came together in only one place, though he is frozen before he can continue. This one place is actually on the Declaration of the Rights of Man and of the Citizen of France, which was approved in 1789. Coincidentally, 89-17=72.
*An arch on a bridge in the famous Leonardo da Vinci painting, the Mona Lisa, seems to have the number 72.
*In Assassin's Creed: Revelations, Ezio can train a total of 72 recruits to the rank of Assassin, not including Rhodes.
*In Leonardo Da Vinci's sketch "Vitruvian Man", the space between the man's lower arm to upper leg has an angle of 72 degrees.
*Desmond Miles was born on the 72nd day of the year 1987.
}} The Conflict with Şehzade Ahmet was an event in 1512, in which Assassin Ezio Auditore, assassinated Ahmet with the help of Selim I.

Constantinople
Subsequently after the assassination of Manuel Palaiologos, Ezio discoveries Ahmet was leading the underground expedition in Cappadocia as the master of the South-East Europe Templars claiming to unite the Ottoman family of Constantinople together with the immeasurably powerful object inside the interior of Masyaf. In his ambition for Ezio to provide the keys for him to open the library, he claimed that if he rejects the offer, he will murder Sofia Sartor.

Arriving at port of Constantinople in 1512, Ezio overhears a herald from a request for the Ottoman about the loads of money a civilian will get for returning Ezio to the authorities. Upon hearing this, Ezio suspects that Ahmet had may not or have kidnapped Sofia yet, decides to travel to her shop to investigate. He finds at the place trashed with multiple died guard bodies on the ground and Sofia kidnapped from her shop. By employing the use of his Eagle Vision, Ezio finds Yusuf Tazim lying died on a bench at the fourth room of her shop. Pierce through his body was a dagger attached to note, suspecting that he did this to protect Sofia. The surrounding Assassin decided to assist Ezio to Arsenal, as it said on the note to do so.
Arsenal Infiltration
Ezio assassinates the surrounding Byzantine guards by himself or his Assassins in the Arsenal Port, leading him to Ahmet hiding in the central fortress. He converses with him about his treachery as a Templar, relying that he was only trying to bring peace. Not in a position to negotiate, Ezio decides to trade with Ahmet; the five keys for Sofia Sartor.
Galata Tower
Ezio along with Assassin approaches Ahmet accompanied with two other Byzantines brings him the five keys in a red pouch at the boarder of Constantinople. Once they trade, Ahmet shows that Sofia on top the Galata Tower with her head trapped in a bag getting coked by a guard. Once Ezio is able to climb up the Galata Tower he assassinates the guard, seizing the bag covering Sofia’s face he discoveries that it was imposter. He sees that the real Sofia was on ground level getting coked by a guard, suspecting that it would take too long to run there, he parachutes off the structure.
On a Carriage
Ezio arrives at Sofia assassinating the guard telling her to get on a carriage, taking the carriage to stop Ahmet from keeping the keys. As he does, Ezio rams Ahmet carriage, weakening his vehicle. The rope attaching the other part of the carriage breaks, leaving Ezio hanging on the rope. He parachutes off the rope kill multiple horsemen with his hook blade and then using his parachute to get back up again. After a long trip through the country side they reenter the city side, as Sofia drives Ezio’s carriage. Ezio jumps down on Ahmet making the two drop down a cliff. As they do, Ezio weakens Ahmet tremendously. When they land, Salim I appears with a large army, choking Ahmet breaking the fencing, letting him plummet down to his death.
Aftermath
After discovering retrieving the all the keys, Ezio immediately set off for the great city of Masyaf in search of the hidden library.
A New Regime is a memory in Assassin's Creed: Revelations
Description
Altaïr, now 64, returns to Masyaf with his wife, Maria Thorpe, after a ten year absence to meet with Abbas Sofian. He wants to know why his youngest son was killed. Abbas reveals that he wants the Apple for himself. Altaïr shows the Apple and demands answers. Swami approaches and puts his hand on the Apple and then reveals details about Sef's death. Altaïr uses the Apple's power on Swami, who manages to stab Maria. Altaïr and Darim are forced to flee Masyaf.

Dialogue
Altaïr and Maria speak with eachother while making their way to the Assassin fortress.
*Altaïr: The Templars have retaken their archive on Cyprus. Abbas sent no reinforcements. It was a massacre.
*Maria: Oh my God...
*Altaïr: Maria, when we left Masyaf ten years ago, this Order was strong. But all our progress has been undone.
*Maria: Abbas must answer for this.
*Altaïr: Answer to whom? The Assassins obey only his command now.
*Maria: Resist your desire for revenge, Altaïr. Speak truth and they will see their error.
*Altaïr: He executed our youngest son, Maria! He deserves to die!
*Maria: Perhaps. But if you cannot win back the Order by honorable means, its foundation will crumble.
*Altaïr: You are right. Thirty years ago I let passion overtake my reason. And it caused a rift that has never fully healed.
*Maria: Speak reasonably, and reasonable men will listen.
*Altaïr: Some will, but not Abbas. I should have expelled him thirty years ago when he tried to steal the Apple.
*Maria: But you earned the respect of the other Assassins because you let him stay.
*Altaïr: How do you know this? You were not there.
*Maria: I married a masterful storyteller.
They enter the Assassin fortress.
*Altaïr: Look at this place. Masyaf is a shadow of its former self.
*Maria: We have been away for a long time.
*Altaïr: But not in hiding. The Mongol threat demanded our attention, and we rode to meet it. What man here can say the same?
*Maria: Where is our eldest son? Does Darim know his brother is dead?
*Altaïr: I sent him a message four days ago. With luck it has reached him already.<br />Abbas... I almost pity him. He wears his grudge like a cloak.
*Maria: His wound is deep. It will help him to hear the truth.
*Altaïr: As I walk these streets, I sense a great fear in the people, not love.
*Maria: Abbas has dismantled this place, and robbed it of all joy.
*Altaïr: We may be walking to our doom, Maria.
*Maria: We may. But we walk together.
Altaïr and Maria confront Abbas.
*Abbas: Let them speak.
*Altaïr: We seek the truth about our son's death. Why was Sef killed?
*Abbas: Is it the truth you want, or an excuse for revenge?
*Maria: If the truth gives us an excuse, we will act on it.
*Abbas: Surrender the Apple, Altaïr, and I will tell you why your son was put to death.
*Altaïr: Ah, the truth is out already! Abbas wants the Apple for himself. Not to open your minds, but to control them!
*Abbas: You have held that artifact for thirty years, Altaïr, reveling in its power and hoarding its secrets. It has corrupted you.
*Altaïr: Very well, Abbas. Take it.
*Maria: What?
Swami walks up to Altaïr and places his hand on the Apple.
*Swami: Before I executed your son, I told him you ordered it yourself. He died believing you had betrayed him.
Altaïr activates the Apple's power.
*Swami: Ah!
*Maria: Altaïr!<br />Altaïr, no!
Swami stabs Maria, and Altaïr stabs Swami.
*Maria: Strength, Altaïr.
*Altaïr: Maria...
*Abbas: He is possessed! Kill him! Take the Apple! Now!
Altaïr makes his way out of the fortress and catches up with his son, Darim.
*Darim: Father! I got your message. What has happened?
*Altaïr: Darim! Turn back!
*Darim: Have they all gone mad?
*Altaïr: We have to go. Abbas must not get his hands on the Apple.
*Darim: If you need more knives, take them.
Altaïr and Darim make their way out of the village, killing the attacking Assassins.
*Darim: Where is mother?
*Altaïr: She is gone, Darim. I'm sorry...
*Darim: What? How?!
*Altaïr: Later. First we fight.
*Darim: Brothers! We need not fight.
*Altaïr: Forget reason, Darim. They have been poisoned by lies.
*Darim: Was it Abbas who killed my brother?
*Altaïr: He killed your brother, Malik, and countless others.
*Darim: He is a madman.
*Altaïr: A madman with an army.
*Darim: He will die. One day he will pay.
*Altaïr: It is time to go!
Altaïr and Darim leave the village of Masyaf.
*Abbas: I will have the Apple, Altaïr! And I will have your head for all the dishonor you brought upon my family! You cannot run forever! Not from us, and not from your lies!
*Altaïr: Maria. My love...

Outcome
Altaïr and Darim flee Masyaf after the death of Maria. Masyaf, and the Assassin Order, are left in Abbas' hands.
