Subject 16 was an individual kidnapped by Abstergo Industries and forced into the Animus before Desmond Miles. A descendant of Ezio Auditore da Firenze, he is related to Desmond Miles, as indicated by portions of their DNA matching in Assassin's Creed II. It is also revealed that he had an ancestor at the Battle of Gettysburg, in the Roman forum and connected with a Queen Isabella.

Biography
Life at Abstergo
The individual known only as Subject 16 was captured by the Templar-front organization Abstergo Industries at some point in 2012 for use in the genetic memory recall program. Kept within the Abstergo Laboratory at all times by Warren Vidic, 16 was kept connected to the Animus for dangerously long periods, sometimes even days at a time. This led to 16 suffering from a severe condition known as the "bleeding effect".

Suffering the bleeding effect, 16 began to explore the memories of his ancestors without the use of the Animus , however his inability to actually control these visions forced Subject 16 into a downward spiral of insanity, eventually culminating in his suicide.

Before his death, 16 was forced by Abstergo to explore the memories of an ancestor named Ezio Auditore da Firenze, who the Templars knew had located the Vault during his lifetime; 16 never relived the memories of his ancestor visiting the Vault. Draining his body of its blood to paint several cryptic messages upon the floor and walls of the Abstergo Laboratory, 16 committed suicide to prevent the Templars reaching their goals.

Legacy
Following his death, the Templars were left without the location of the Vault, but the knowledge that an ancestor of Ezio’s – Altaïr Ibn-La'Ahad – had once held a Piece of Eden. Knowing this, Abstergo kidnapped yet another descendant of Ezio and Altaïr, Desmond Miles. During his incarceration, Desmond spoke often with Dr. Vidic’s assistant Lucy Stillman about many things, including his predecessor; accessing private e-mails, he learned even more. Later still, following Desmond’s escape from Abstergo and the beginning of his journeys through the memories of Ezio Auditore the Assassins learned that Subject 16 had managed to hack the Abstergo Animus before his death, and placed twenty encrypted files, called glyphs, throughout the memories. These hacks made their way on to Lucy's portable device when she used it on the Abstergo Animus in order to gain details and memory sequences on Ezio Auditore da Firenze. Eventually the Glyph hacks were transferred unknowingly onto Rebecca Crane's Animus 2.0.

After decrypting all twenty files, the assassins were rewarded with a genetic memory dated "date classified BCE". The video, shows two free-runners climbing a large glass structure, Piece of Eden in hand; before referring to one another as Adam and Eve.
Subject 12 was the direct descendent of an individual involved in (or subjected to) the Philadelphia Experiment, a popular conspiracy theory where a naval vessel is alleged to have travelled foward in time for 18 minutes. The email claims that the Templars obtained sufficient information to reconstruct the device responsible, but will not continue with development due to concerns about the creation of irreparable time paradoxes.

Subject 16
Subject 16
Subject Sixteen was the predecessor to Desmond Miles in Abstergo's experiments. His name is unknown, but he is at one point referred to as "Neumann". He is never seen in any game.

It is known that, due to the "Bleeding effect", Subject 16 went mad and wrote the cryptic messages throughout the Animus room and his bedroom, using his own blood (dying afterwards). He may also have, in his madness, murdered Leila Marino and used her blood to write some of the cryptic messages as it is even stated by Subject 16 himself that if he would have written all of the messages with his own blood, he would have died before he finished.

Subject 17 (Desmond Miles)
Desmond Miles
Desmond Miles is the true protagonist of Assassin's Creed, and the successor of Subject 16.

Brought to Abstergo against his will, he was threatened with a coma and death if he did not participate in the Animus project. Giving in to the threat, he entered the Animus. He was made to recreate the actions of his ancestor Altaïr, in order to find the location of the Piece of Eden. Like Subject 16 before him, Desmond experiences the Bleeding effect. However, he only experiences a relatively minor change- the ability to use Eagle Vision like his ancestor. This is probably due to Lucy urging him to take breaks from the Animus. In Assassins Creed 2 though he gains all of Ezio's skills. In Assassin's Creed Abstergo were about to kill him until Lucy suggested that he can still be useful for finding more Pieces of Eden.

Subject ? (Daniel Cross)
Daniel Cross
Daniel Cross was a recovering alcoholic and drug user who was brought to Abstergo to relive the memories of his ancestor, Nikolai Orelov. Not much is known about Daniel at this point.

Assassin's Creed: Brotherhood
It has been noticed in the multiplayer walkthrough video that the players in the game are referred to as "Subjects" e.g. "Subject 6 killed Subject 2". This could likely be replaced with player's gamertags when Assassin's Creed: Brotherhood goes live.
