Subject 16 is an individual that Abstergo Industries kidnapped and forced in the Animus before Desmond Miles ("Subject 17") and another descendant of Altaïr Ibn-La'Ahad and Ezio Auditore da Firenze. He is related to Desmond, as portions of their DNA matched in Assassin's Creed II, and it is also revealed that he had an ancestor at the Battle of Gettysburg, in the Roman forum and one connected with a Queen Isabella.

Assassin's Creed
Former Abstergo employee and researcher Lucy reveals in one of her e-mails to her supervisor, Warren Vidic, that Subject 16 has Far Eastern origins, as one of his ancestors was "involved in an important event" in said area. A different e-mail possibly indicates that his name is "Neumann". The same email also hints that former employee (now deceased) Leila Marino of Abstergo Industries may have been romantically involved with Subject 16. This suggests a romantic involvement prior to his kidnapping or that Leila worked with Subject 16 in the same way as Lucy works with Desmond.

At Abstergo's orders, Subject 16 was kept connected to the Animus for dangerously long sessions, sometimes even days at a time. This lead to a severe and powerful "bleeding effect" as Subject 16 became more capable of revisiting these genetic memories without the use of the Animus. Due to his inability to actually control these phenomena, Subject 16 slowly spiraled into insanity, eventually culminating in suicide. He describes said events in the glyphs. "I'm holding a rifle, Gettysburg is in the distance, I just stabbed a man, there's blood on my waistcoat." Not much else is known about Subject 16.

At the end of Assassin's Creed, due to his own experience with the bleeding effect, Desmond is able to use Eagle Vision to view several cryptic messages that were drawn in blood. Subject 16 states in one message that he drained his own body to paint the figures on the floor and walls of the main room where the Animus is located. But if he had used his own blood to paint the wall mural in the sleeping room, he would have bled out — literally — before he finished his work. It is possible, but not probable that some of the messages were drawn with Leila Marino's blood and, in part, the cause of her death.

The symbols painted on the walls and floors later reappear in Assassin's Creed II as glyphs, parts of encrypted files that Subject 16 added to genetic memories after he hacked the Animus.

Assassin's Creed II
Subject 16 makes his first vocal appearance in Assassin's Creed II, voicing-over twenty encrypted files that he hacked from Abstergo's mainframe. After all twenty files are decrypted, they combine to form a short video which contains what Subject 16 claims to be "The Truth" about the origins of humanity. In the video, a man and a woman who refer to one another as Adam and Eve race through a modern complex, with Eve carrying "the apple" (which is, in fact, one of the Pieces of Eden). The short diatribes from these files, as well as commentary from Lucy, reveal a bit more about the extent of the madness which lead to Subject 16's death. In his final commentary in the twentieth file, Subject 16 only states, "Lucy, I can't wait any longer. I am ready to go now... She sees me raise the knife."

In the beginning of the game, Lucy searches for the closest genetic match between Subject 16 and Desmond, and the memory of Ezio's birth appears.

Description
Brutes are enemies featured in Assassin's Creed II. They are essentially human tanks, wearing heavy, durable armor and carrying large, heavy weapons, and also moving very slowly due to the weight of their equipment. They are the strongest guards in terms of combat difficulty in the game, but can easily be avoided.

Combat
Brutes are tricky enemies to defeat because of the nature of their special attacks and normal attacks.

*Normal attacks: Brute normal attacks can be blocked with any weapon but can't be countered with a short blade or sheathable weapon. They can however, be countered (lethally) with another heavy weapon such as those the Brutes themselves carry, a long weapon (carried by Seekers), or by the Hidden Blade(s), although the successful timing window for the latter is rather short (a fraction of a second). They can also be countered with Fists, which will disarm the Brute which, followed up by a quick attack on the unarmed Brute, can kill him quickly, but the timing for the disarm is short, although apparently slightly longer than for the Hidden Blade(s). Special care should be taken when trying to counter the Brutes' with the Hidden Blade(s), as, because of the very short timing window for successful counter kills, it is highly possible to fail a counter-kill. This failure is penalized severely, as Brute attacks deal great amounts of damage and, should the Brute perform a combo, you may lose a significant chunk of your health bar. A less risky but more time consuming tactic is to dodge the Brute's attacks and follow up with a combo of your own. Performed correctly, the combo may end with an execution. The maximum number of hits in a Brute combo is three.
*Special attacks: Neither of the Brutes' special attacks can be blocked or countered. It is suggested that you either attack the Brute before he actually executes the attack, dodge and fire back with an attack of your own, or simply walk out of range of the attack (since the Brute, after beginning to charge a special attack, cannot move again until after performing it) and, again, attack back.
#Smash: The Brute takes a strong downward swing with his weapon, knocking Ezio on his back and doing about as much damage as a normal attack. A Brute is about to perform a smash attack if he raises his weapon high above his head. A Smash special attack takes less time for a Brute to prepare (approx. one second) than a Knock Weapon Away special attack.
#Knock Weapon Away: This special attack is not named 'Disarm' because it is not the same as Ezio's disarm ability; that is, the Brute does not take Ezio's weapon and use it for himself. Instead, the Brute takes a hard right-to-left swing with his weapon, breaking Ezio's guard for a second and, if Ezio was using a short blade, sheathable weapon or picked up weapon, knocks it out of his hand. The attack does no damage, but, as said above, breaks Ezio's guard for a second, opening him to attacks from other foes in the fight. A Brute is about to perform a Knock Weapon Away special attack if he holds his weapon out behind him. A Knock Weapon Away special attack takes a Brute longer to prepare than a Smash attack, which means a larger window of time to attack them than for a Smash special attack. Note that after Ezio's weapon has been knock away by Brute, the first sheathable weapon Ezio pick up will become his primary weapon.

*Attacking: The best times to attack a Brute are after disarming him, while he is preparing for a special attack, after dodging an attack, or after moving out of range of a special attack.
*Tip: An easy way to kill a brute while taking a large group of guards with him is to drop a smoke bomb and start assassinating the Brutes, they will be to busy choking to counter your attacks.

Intelligence, Agility, and Methods of Escape
Brutes are not highly intelligent and can be escaped once you've gotten out of sight by blending or hiding. They are also not agile or quick in any sense of the words, and cannot free-run or climb (at least not without possible glitching, and even then only for a second or less), so taking to the roofs is an easy method of escape.

Ranking
If someone could please add images and information on Brute rankings here, that would be helpful.

Brutes, like many other guards, have a very high morale. In fact, the Brutes have the highest morale over any guard in Assassin's Creed 2. This means that unlike a few of their allies, they will always fight to the death.

Special Brutes
*One of Ezio's assassination targets, Dante Moro, carries what may be a Bearded Axe with an enlarged head, and fights like a Brute, only with far more health and immunity from being disarmed.

*One of Ezio's allies, Bartolomeo, carries what appears to be a Bastard Sword, (although the design and coloring of the handle appears different from those carried by Brute guards) which he calls "Bianca" and prizes, and behaves like a Brute in combat, only with far more health.

*Mario Auditore carries a Bearded Axe and attacks like a brute
